Question: How to generate pileup file

I am doing research on variant identification of genetic disease. when i generate file frome pileup tool in galaxy then it cannot created but when i use mpileup tool then this line is shown
[mpileup] 1 samples in 1 input files
<mpileup> Set max per-file depth to 8000
Now i am stuk on this point i cannot generate pileup file. If you help me out then i will be very thankful to you.
